Use the Google or Yahoo Geocoding/Maps APIs.
I implemented a store locator for a client where you can enter a postcode, suburb or full address into a text box, and it will ask Google via their API to find its location, and then find the distance from that location to all of the stores, returning the top n results. I did find that it works much better if you append " Australia" to whatever they type into the text box though.
I used the GeoKit plugin for Rails, it made things stupidly easy.
